Deletion of individual Robo1 Ig2-5 domains does not interfere with Slit binding in cultured Drosophila cells. Drosophila S2R+ cells were transfected with the indicated HA-tagged UAS-Robo1 transgenes, and treated with conditioned media from cells expressing Slit. After Slit treatment, cells were fixed and stained with anti-HA (magenta) to detect expression of Robo1 variants, and anti-Slit (green). Slit binds robustly to cells expressing full-length Robo1 (b), but not to mock-transfected cells (a) or cells expressing Robo1∆Ig1 (c). Cells expressing Robo1∆Ig2 (d), Robo1∆Ig3 (e), Robo1∆Ig4 (f), or Robo1∆Ig5 (g) exhibit a similar level of Slit binding to cells expressing full-length Robo1.
