Morphological characterization of RGCs from RetCreERt2/WT; Brn3aKO/WT; ROSA26AP/WT and RetCreERt2/WT; Brn3aWT/WT; ROSA26AP/WT retinas. Cre was activated by i.p. administration of 250 μg 4-HT to mothers of the pups at gestational day 15 (E15 for pups), and RGCs visualized by recombination of the ROSA26AP reporter. Labelled RGCs have the same Ret and Brn3a gene copy numbers as their respective retinas (either RetKO/WT; Brn3aKO/WT—left panels, or RetKO/WT; Brn3aWT/WT—right panels). RGCs were labelled in retinas from RetCreERt2/WT; ROSA26AP/WT; Brn3aKO/WT (gray, n = 169 cells from 6 mice, derived from 3 distinct experiments/litters) and RetCreERt2/WT; ROSA26AP/WT; Brn3aWT/WT (white, n = 138 cells from 3 mice, derived from 2 distinct experiments/litters) mice. a Retinal flat mounts from RetCreERt2/WT; Brn3aKO/WT; ROSA26AP/WT (left) and RetCreERt2/WT; Brn3aWT/WT; ROSA26AP/WT (right) mice, processed for AP histochemistry.
